This kit contains PCR mixes and reagents for laboratory detection of Yersinia pestis and Francisella tularensis RNA with electrophoretic evaluation.

PCR Mixes and Reagents

1. PCR Mix 1 “Y. pestis”
   Transparent, colorless solution containing:
   • Mixture of two primers (“Sintol”, Russia), each at 8 pmol
   • Deoxynucleotide triphosphates (dNTPs: dATP, dGTP, dCTP, dTTP) (“SibEnzyme”, Russia), each at 1 mM
   • Sodium azide (“Sigma”, USA, or equivalent) - 0.031%
2. PCR Mix 1 “F. tularensis”
   Transparent, colorless solution containing:
   • Mixture of two primers (“Sintol”, Russia), each at 8 pmol
   • Deoxynucleotide triphosphates (dATP, dGTP, dCTP, dTTP) (“SibEnzyme”, Russia), each at 1 mM
   • Sodium azide (“Sigma”, USA, or equivalent) - 0.031%
3. PCR Mix 2
   Transparent, colorless solution containing:
   • Tris-HCl - 167.5 mM, pH 8.8 at 25 °C (“SibEnzyme”, Russia)
   • Ammonium sulfate (“SibEnzyme”, Russia) - 41.5 mM
   • Magnesium chloride (“SibEnzyme”, Russia) - 5.0 mM
   • Tween-20 (“SibEnzyme”, Russia) - 0.025%
4. Taq Polymerase
   Transparent, colorless solution (“SibEnzyme”, Russia) - 5 U/µl
5. TE Buffer
   Transparent, colorless solution containing:
   • Tris-HCl (“Amresco”, USA) - 10 mM
   • Na2-EDTA (“Amresco”, USA) - 1 mM
6. RNase-free water, transparent, colorless (“Fermentas”, USA)
7. Mineral oil, colorless viscous liquid (light white paraffin oil, “AppliChem”, USA)
8. DNase, transparent, colorless solution (“Promega”, USA) - 1 U/µl
9. DNase Reaction Buffer
   Transparent, colorless solution containing:
   • Tris-HCl, pH 8.0 (“Promega”, USA) - 400 mM
   • Magnesium sulfate (“Promega”, USA) - 100 mM
   • Calcium chloride (“Promega”, USA) - 10 mM
10. DNase Stop Buffer
    Transparent, colorless solution containing EGTA, pH 8.0 (“Promega”, USA) - 20 mM
11. Positive Control DNA Sample for Y. pestis
    Transparent, colorless aqueous solution containing 1 ng of Y. pestis DNA
12. Positive Control DNA Sample for F. tularensis
    Transparent, colorless aqueous solution containing 1 ng of F. tularensis DNA
