This reagent kit is designed for the detection of Yersinia pestis DNA using the polymerase chain reaction (PCR) method.

Kit Composition

PCR Mix 1

A clear, colorless solution containing:

Mixture of two primers ("Sintol", Russia), each at a concentration of 8 pmol;
Probe ("Sintol", Russia) at a concentration of 4 pmol;
Mixture of deoxynucleoside triphosphates (dNTPs) – dATP, dGTP, dCTP, dTTP ("SibEnzim", Russia), each at 1 mM;
Sodium azide ("Sigma", USA, or equivalent) – 0.031%.

PCR Mix 2

A clear, colorless solution containing:

Tris-HCl – 167.5 mM, pH 8.8 at 25°C ("SibEnzim", Russia);
Ammonium sulfate ("SibEnzim", Russia) – 41.5 mM;
Magnesium chloride ("SibEnzim", Russia) – 5.0 mM;
Tween-20 ("SibEnzim", Russia) – 0.025%.

Taq Polymerase

Clear, colorless solution ("SibEnzim", Russia) – 5 units/µL.

TE Buffer

Clear, colorless solution containing:

Tris-HCl ("Amresco", USA) – 10 mM;
Na2-EDTA ("Amresco", USA) – 1 mM.

Positive Control DNA Sample (Y. pestis DNA)

Clear, colorless solution containing an aqueous solution of Yersinia pestis DNA at a concentration of 1 ng.
